Research progress of lactylation in tumor evolution

Abstract

The enhanced glycolysis of tumor cells due to metabolic reprogramming gives rise to a large accumulation of lactic acid, which creates conditions for lactylation. As one new type of protein post-translational modification, the lactylation’s role in tumor evolution has become a current research hotspot. Lactylation can profoundly affect Tumorigenesis and development of tumors via regulating tumor cells’ metabolic reprogramming, DNA damage repair, immune microenvironment, and so on. In-depth study of lactylation’s role in tumor development will not only help to reveal the molecular mechanism of tumor evolution, but also provide new ideas and strategies for early diagnosis, therapy and prognosis evaluation of tumors. This article summarizes the advanced and classical understanding of lactate and lactylation, and provides a comprehensive literature review of lactylation in cancer stem cells and cancer, designing to summarize and analyze the research status, trends and key issues of lactylation in the field of cancer.
